I'm an derivative and I-485 is pending on EB, I was working using my EAD and got laid off, i applied for Unemployment Benefits and haven't submitted for any claim so far. Is it ok to go ahead and request for claim or will that impact my pending Green card I-485 application?

You would need to ahve the number of quarters necessary under the law within the jurisdiction where you worked. U/C benefits is a form of insurance nd is not included in the public charge restrictions of the INA.

Unemployment benefits are part of an INSURANCE program ... if you qualify ... you can collect. But don't try to get food stamps.
