There are times when the D.A. will agree to dismiss it after you show that you addressed the issue immediately after learning about your err. There is no guarantee, but I have seen that before.

If the cop is willing to dismiss the ticket upon registration he will usually give you the option during the stop. There is no actual defense to this if you were not registered, there is no notice requirement. If you register immediately and show up to the hearing with proof you can ask for leniency but you violated and there is no "defense" to this violation unless it was actually inspected.
I don't practice law in Pennsylvania, but here in Missouri, you can get a "Failure to Register Vehicle" or "Expired Registration" ticket dismissed upon payment of court costs if you provide the court with proof that you fixed the problem by getting your car registered. In some courts, you are allowed to bring proof of registration to the court clerk prior to the court date; in other courts, you will either have to hire an attorney to take care of it for you or go to court yourself and show proof of registration to the judge. Call the court clerk and ask what their policy is.

I think each jurisdiction is different and a local criminal defense attorney may provide better advice to that effect.
From the above it appears that the infraction was time framed, strict liability to have registration by a certain point and I am doubtful that there is any defense to that effect unless there was a notice issue or some mistake on the notification or agency part.
